DNA Sensing in Mismatch Repair-Deficient Tumor Cells Is Essential for Anti-tumor Immunity

Changzheng Lu et al.

Summary: This study demonstrates the importance of DNA sensing within tumor cells in triggering anti-tumor immunity in dMLH1 tumors, providing new mechanisms and biomarkers for anti-dMMR-cancer immunotherapy.

MLH1 Deficiency-Triggered DNA Hyperexcision by Exonuclease 1 Activates the cGAS-STING Pathway

Junhong Guan et al.

Summary: This study revealed that loss of MutLa subunit MLH1 leads to production of neoantigens and activation of the cGAS-STING pathway, affecting DNA repair process and ultimately causing chromosomal abnormalities and DNA release. This unknown MMR mechanism has significant implications for genome stability and cancer therapy.

ITGBL1 is a new immunomodulator that favors development of melanoma tumors by inhibiting natural killer cells cytotoxicity

Yann Cheli et al.

Summary: Resistance to immunotherapies in melanoma patients is associated with a decrease in cell differentiation and an increase in the mesenchymal phenotype. The loss of MITF plays a role in melanoma cells evading the immune system, with ITGBL1 identified as a key immunomodulator. Inhibiting ITGBL1 could potentially improve melanoma response to immunotherapies.
