Genome-Wide Histone Modifications and CTCF Enrichment Predict Gene Expression in Sheep Macrophages

Alisha T. Massa et al.

Summary: This study conducted the first genome-wide survey of regulatory elements in sheep immune cells, particularly alveolar macrophages, to identify cis-regulatory DNA elements and chromatin domain boundaries controlling gene expression. The findings revealed a large number of reproducible regulatory elements with potential biological functions, exceeding standards set by FAANG and ENCODE. Active elements were predictive of gene expression in alveolar macrophages, while silencer elements were enriched for repressed developmental genes, demonstrating the importance of regulatory elements in gene control mechanisms.

Genome-wide fine-mapping identifies pleiotropic and functional variants that predict many traits across global cattle populations

Ruidong Xiang et al.

Summary: This study presents a method for fine-mapping potentially causal mutations by integrating functional, evolutionary, and pleiotropic information and developing a custom genotyping array. The custom array, containing variants with biological functions, outperformed the standard array in predicting genetic values for multiple traits in dairy cattle populations across different countries. This highlights the potential improvement in genomic prediction by utilizing mutations with functional, evolutionary, and pleiotropic consequences.
