This suggests HOXB-AS3 could be a predictor of OS and a potential target for EOC therapies.

Aerobic glycolysis in TNBC not only promotes aggressiveness of tumor cells but also initiates a positive regulatory loop for enhancing tumor progression by modulating the inflammatory TME.

In CMM, LINC00518 induces radioresistance by regulating the glycolysis pathway through a negative feedback loop involving miR-33a-3p and hypoxia-inducible factor 1 alpha (HIF-1 alpha). Knockdown of LINC00518 expression sensitizes CMM cells to radiotherapy, suggesting a potential strategy to improve treatment outcomes.

Development of antitumor biguanides targeting energy metabolism and stress responses in the tumor microenvironment

Summary: The development of potent new biguanide derivatives through structural modification of the arylbiguanide scaffold resulted in compounds with strong inhibitory effects on HIF-1 and UPR-mediated transcriptional activation, as well as selective cytotoxicity towards cancer cells under low glucose conditions. These biguanides were found to strongly inhibit oxidative phosphorylation and activate compensative glycolysis in the presence of glucose, while suppressing both processes in the absence of glucose, leading to cellular energy depletion and apoptosis. This data suggests that the pleiotropic effects of these biguanides could provide a more targeted and effective treatment for cancer cells by suppressing stress adaptation systems in the tumor microenvironment.
