Conversion of T Follicular Helper Cells to T Follicular Regulatory Cells by Interleukin-2 Through Transcriptional Regulation in Systemic Lupus Erythematosus

Summary: In patients with SLE, the proportion of Tfh cells increased, while the activated Tfr cells decreased. IL-2 induced the conversion of memory Tfh cells to functional Tfr cells by binding to FOXP3 and BCL6 gene loci accompanied by suppression of H3K27me3. IL-2 stimulation suppressed the generation of CD38+CD27 high plasmablasts in Tfh and B cell coculture assays.

Summary: The study found that in individuals with inflammatory bowel disease, there is a progressive expansion of ROR gamma t(+) T-reg cells, and activating Wnt-beta-catenin signaling can induce the expression of pro-inflammatory cytokines and the ROR gamma t(+) T-reg phenotype in these cells. TCF-1 and Foxp3 together limit the expression of pro-inflammatory genes in T-reg cells.
