The extracellular matrix: A key player in the pathogenesis of hematologic malignancies

Ishnoor Sidhu et al.

Summary: This article discusses the importance of hematopoietic stem cells and the bone marrow microenvironment in hematologic malignancies, as well as the impact of genetic mutations in this process. It also mentions the role of the extracellular matrix in promoting the evolution of hematologic cancers, although its precise role is not fully understood.

Vascular Notch Signaling in Stress Hematopoiesis

Can Huang et al.

Summary: Canonical Notch signaling is crucial for cell proliferation, differentiation, and fate maintenance in development and cancer, with distinctive roles in hematopoietic stem cell emergence during embryonic development and lineage choices in adult hematopoiesis. Studies continue to debate the necessity of Notch signaling for adult HSC maintenance, urging for more detailed investigations into specific biological contexts to better understand the therapeutic implications of Notch-related pathways.

Deficiency of Rbpj Leads to Defective Stress-Induced Hematopoietic Stem Cell Functions and Hif Mediated Activation of Non-canonical Notch Signaling Pathways

Ram Lakhan et al.

Summary: Deletion of canonical notch pathways in hematopoietic stem cells leads to compromised HSC maintenance and functions in response to stressors, while upregulating key notch target genes and increasing notch activity, which establish a functional link between Hif and Notch pathways in hematopoiesis.

A mechanosensitive peri-arteriolar niche for osteogenesis and lymphopoiesis

Bo Shen et al.

Summary: Stromal cells in adult bone marrow expressing leptin receptor are a critical source of growth factors for haematopoietic stem cells. Among these cells, those expressing osteogenic growth factor osteolectin are poised for osteogenesis, while those near sinusoids are primed for adipogenesis. Mechanical stimulation is crucial for the maintenance of the peri-arteriolar niche for osteogenesis and lymphopoiesis.

Decline in IGF1 in the bone marrow microenvironment initiates hematopoietic stem cell aging

Kira Young et al.

Summary: As individuals age, the function of hematopoietic stem cells (HSCs) declines, leading to limited health span of the blood and immune systems. Research indicates that the hallmarks of aging in HSCs and hematopoiesis start accumulating by middle age, with decreased levels of the longevity-associated molecule IGF1 in the bone marrow microenvironment playing a key role in causing HSC aging. By directly stimulating middle-aged HSCs with IGF1, molecular and functional hallmarks of aging can be rescued, highlighting the complex relationship between IGF1 levels, longevity, and HSC function.

Targeting chemokines for acute lymphoblastic leukemia therapy

Zixi Hong et al.

Summary: Acute lymphoblastic leukemia (ALL) is regulated by various signaling molecules such as cytokines and adhesion molecules in its microenvironment. Chemokines play important roles in leukemia microenvironment and affect ALL outcomes. Targeting chemokine axes for ALL treatments shows potential, with some related inhibitors entering clinical trials.
