The order and logic of CD4 versus CD8 lineage choice and differentiation in mouse thymus

Summary: In this study, the authors used single cell RNA sequencing to analyze coreceptor gene expression in mouse thymocyte selection intermediates. They found that the timing of coreceptor expression is flexible depending on TCR signal strength, and that selection intermediates can discern MHC class before losing coreceptor expression. This suggests a model where signal strength influences the timing of coreceptor gene activity, ultimately impacting CD4/CD8 lineage choice in developing T cells.

Differentiation programs for CD4+ T-FH and CD8+ T-SCM cells are compared and contrasted with their effector counterparts, T-H1 and T-SLEC cells, to understand T cell differentiation and effector function.
