An Overview of Proteases: Production, Downstream Processes and Industrial Applications

Recombinant neutral protease rNpI as fish feed additive to improve protein digestion and growth

Summary: The study found that the neutral protease rNpI effectively hydrolyzed soybean protein isolates, resulting in a high degree of hydrolysis and small-peptide yield, with notable antioxidant activity for fish health. rNpI complemented trypsin in soybean protein digestion better than papain or Alcalase. The addition of rNpI led to lower feed protein and energy content, reduced FCR, and improved growth performance in fish, with the best results seen in fish fed a diet with reduced rapeseed meal and wheat content.

The recombinantly produced ASP demonstrated remarkable pH stability, notable thermostability, and high specific activity at extreme alkaline conditions, making it a potential novel protease candidate for various industrial sectors such as textile, detergent, leather, feed, waste, pharmaceutical, and others.

Summary: The study focused on characterizing proteases mined from solid tannery waste metagenome, including physico-chemical analysis, secondary structure analysis, conservation analysis, diversity analysis, and homology modeling. The results showed the presence of conserved motifs in the proteases and successful molecular docking with specific peptide ligands, indicating potential catalytic activity for hydrolyzing peptide substrates. This structural and functional insight may lead to the discovery of novel industrially important proteases.

Functional screen of (meta)genomic libraries can potentially identify new enzymes, but discovering enzymes that meet industry compliance demands remains challenging, especially in the case of proteases. This study demonstrated the use of six industry-relevant animal and plant by-products as complementary substrates in functional screens, suggesting temperature as a screening parameter to potentially discover new broad-substrate range and robust proteases.
