Timing without coding: How do long non-coding RNAs regulate circadian rhythms

Rebecca A. Mosig et al.

Summary: Long non-coding RNAs (lncRNAs) are a type of regulatory RNA that plays important roles in various biological processes. Recent studies have highlighted their significance in the circadian clock system. This review provides an overview of our current understanding of how lncRNAs regulate the circadian clock and discusses the unresolved questions in the field.

Dysregulation of the histone demethylase KDM6B in alcohol dependence is associated with epigenetic regulation of inflammatory signaling pathways

Andrea L. Johnstone et al.

Summary: This study identified 11 dysregulated epigenetic enzymes in the brain of alcohol-dependent rats during protracted abstinence, with KDM6B showing region-specific dysregulation in the prefrontal cortex and nucleus accumbens. The upregulation of KDM6B in alcohol-dependent rats was associated with decreased trimethylation levels at histone H3, lysine 27, and enrichment of alcohol-induced changes in H3K27me3 at genes in the IL-6 signaling pathway, implicating a novel KDM6B-mediated epigenetic signaling pathway integrated with inflammatory pathways in the development of alcohol addiction.

Methionine Supplementation Abolishes Nicotine-Induced Place Preference in Zebrafish: a Behavioral and Molecular Analysis

Antonella Pisera-Fuster et al.

Summary: This study found that oral L-methionine supplementation reduces nicotine-induced conditioned place preference (CPP) and drug reward-related behaviors in zebrafish, possibly by influencing DNA methylation processes. The upregulation of DNA methyltransferases and histone methyltransferases, along with the downregulation of certain genes, were associated with this effect. Overall, these findings suggest that L-methionine may be a potential therapeutic drug for treating nicotine addiction.

A genome-wide microRNA screen identifies the microRNA-183/96/182 cluster as a modulator of circadian rhythms

Lili Zhou et al.

Summary: This study identified the miRNA cluster miR-183/96/182 as key regulators of circadian rhythms, with miR-96 directly targeting the core circadian clock gene PER2. Knockout of the miR-183/96/182 cluster in mice showed tissue-specific effects on circadian parameters, highlighting the importance of miRNAs as a genome-wide layer of circadian clock regulation.

Interaction between cocaine use and sleep behavior: A comprehensive review of cocaine's disrupting influence on sleep behavior and sleep disruptions influence on reward seeking

E. Theresa Bjorness et al.

Summary: Dopamine, orexin, and adenosine systems play dual roles in reward and sleep/arousal. Drug abuse may influence both reward and sleep/arousal mechanisms. Cocaine can cause acute sleep loss and abnormal recovery patterns under chronic exposure conditions. Experimentally induced sleep disturbance may increase cocaine seeking behavior.

Psychostimulant Use Disorder, an Unmet Therapeutic Goal: Can Modafinil Narrow the Gap?

Melinda Hersey et al.

Summary: The rapid increase in individuals affected by psychostimulant use disorder (PSUD) has highlighted the potential of Modafinil (MOD) as a therapeutic option due to its non-addictive properties, despite its similar mechanism of action to commonly abused psychostimulants on the dopamine transporter (DAT). Clinical and preclinical studies have demonstrated the efficacy of MOD in treating symptoms related to PSUD and other psychiatric disorders involving alterations of brain dopamine systems.

Rhythmic Regulation of DNA Methylation Factors and Core-Clock Genes in Brain Structures Activated by Cocaine or Sucrose: Potential Role of Chromatin Remodeling

Lamis Saad et al.

Summary: The study reveals the interaction between the circadian system and the reward system in modulating reward and memory, showing distinct effects of cocaine and sucrose on gene expression and DNA methylation processes in dopamine projection areas interconnected with the reward system. Understanding these mechanisms is crucial for developing selective therapeutic tools for compulsive behaviors.

Summary: The obsessive-compulsive spectrum encompasses disorders with similar core features, such as obsessive thoughts, compulsive behaviors, and anxiety. Ketamine has shown remarkable results in treating depression, PTSD, and OCD, but its efficacy in SUD and ED remains uncertain. Further studies are needed to clarify its potential in these conditions.
