Molecular dynamics analysis predicts ritonavir and naloxegol strongly block the SARS-CoV-2 spike protein-hACE2 binding

Milad Bagheri et al.

Summary: The emergence of COVID-19 has made humans vulnerable to the SARS-CoV-2 virus. Drug repurposing approaches are believed to be helpful in finding treatments. Researchers screened FDA-approved ligands and identified ritonavir and naloxegol as potential therapeutics against COVID-19.

The tyrosine kinase inhibitor nilotinib inhibits SARS-CoV-2 in vitro

Valeria Cagno et al.

Summary: Since the emergence of SARS-CoV-2 in late 2019, there has been no approved vaccine to combat the infection, highlighting the urgent need for effective treatment for the ongoing COVID-19 pandemic. Drug repurposing offers a rapid antiviral response in such emergencies. Nilotinib shows promising activity against SARS-CoV-2 in vitro, making it a potential candidate for COVID-19 treatment in vivo.

Repurposed Antiviral Drugs for Covid-19-Interim WHO Solidarity Trial Results

Summary: The authors of the study reported the interim results of using four repurposed antiviral drugs (remdesivir, hydroxychloroquine, lopinavir, and interferon beta-1a) in hospitalized COVID-19 patients. The study showed that these drug regimens had little or no effect on overall mortality, initiation of ventilation, and duration of hospital stay.

Methylene Blue Inhibits the SARS-CoV-2 Spike-ACE2 Protein-Protein Interaction-a Mechanism that can Contribute to its Antiviral Activity Against COVID-19

Damir Bojadzic et al.

Summary: Research on organic dyes led to the discovery that methylene blue can inhibit the interaction between SARS-CoV-2 spike protein and receptor ACE2, showing potential antiviral activity in preventing virus entry.

Enhanced production of camptothecin by immobilized callus of Ophiorrhiza mungos and a bioinformatic insight into its potential antiviral effect against SARS-CoV-2

Rajesh Mamkulathil Devasia et al.

Summary: This study successfully immobilized tissue culture grown callus of Ophiorrhiza mungos to produce a higher concentration of CPT, and bioinformatically analyzed the binding potential of CPT towards two important proteins of SARS-CoV-2. Docking analysis showed a potential interaction of CPT against the external spike glycoprotein of SARS-CoV-2.

Repurposing of approved drugs with potential to interact with SARS-CoV-2 receptor

Tamim Ahsan et al.

Summary: The study found that ACE1 is more abundantly expressed in human lung tissue compared to ACE2, and they share similarities in the peptidase domains, which may interact with the SARS-CoV-2 spike protein. Additionally, it predicts that approved drugs may interact with ACE1 and ACE2, potentially interfering with the entry of SARS-CoV-2 into host cells.

Drug Repurposing of Itraconazole and Estradiol Benzoate against COVID-19 by Blocking SARS-CoV-2 Spike Protein-Mediated Membrane Fusion

Chan Yang et al.

Summary: The study found that Itraconazole and Estradiol benzoate can inhibit viral entry by targeting the six-helix fusion core of SARS-CoV-2 S protein, presenting them as novel potential therapeutic remedies for COVID-19 treatment. Further research also showed that Itraconazole has broad-spectrum activity targeting 6-HB in the S2 subunit of SARS-CoV and MERS-CoV S protein, suggesting its potential as a pan-coronavirus fusion inhibitor.
