Differential and unique patterns of synaptic miRNA expression in dorsolateral prefrontal cortex of depressed subjects

Yuta Yoshino et al.

Summary: This study examined the role of miRNAs at synapse in major depressive disorder (MDD) by sequencing and analyzing miRNAs in synaptic and total tissue fractions. The findings suggest that altered miRNAs may be involved in synaptic plasticity, nervous system development, and neurogenesis in MDD pathogenesis. Additionally, shifts in expression ratios of certain miRNAs and changes in miRNA processing enzymes were observed in MDD subjects.

Zygotic exposure to venlafaxine disrupts cortisol stress axis activity in multiple generations of zebrafish

William A. Thompson et al.

Summary: The ubiquitous use of antidepressants, such as venlafaxine, has led to increased concentrations of these pharmaceuticals in waterways, impacting neural development in zebrafish embryos. Early developmental exposure to venlafaxine disrupts the functioning of the hypothalamus-pituitary-interrenal axis in zebrafish, affecting cortisol levels and responses to stress across multiple generations. This study suggests that venlafaxine is an endocrine disruptor with potential long-term and generational consequences on stress axis activity.

Identification of microRNA-9 linking the effects of childhood maltreatment on depression using amygdala connectivity

Cancan He et al.

Summary: Childhood maltreatment is considered a significant risk factor for major depressive disorder. This study investigated the neural basis of the effects of childhood maltreatment on depression, with a focus on microRNA-9. Higher miR-9 levels in MDD patients were associated with lower childhood maltreatment scores and less severe depressive symptoms, with overlapping effects on amygdala functional connectivity networks. The study also found that a support vector classifier model incorporating miR-9 levels, childhood maltreatment severity, and AFC strength could accurately differentiate MDD patients from healthy controls.

Distinct epigenetic signatures between adult-onset and late-onset depression

Hirotaka Yamagata et al.

Summary: The study identified DNA methylation markers more suitable for adult-onset depression (AOD) patients than for late-onset depression (LOD) patients. The methylation profile of AOD group was distinct and more homogeneous compared to the LOD group. Six identified methylation CpG sites were validated as potential markers for AOD, and a combination of three specific methylation markers achieved the highest accuracy.

Sex-specific effects of social defeat stress on miRNA expression in the anterior BNST

Pei X. Luo et al.

Summary: Social defeat stress induces sex-specific miRNA expression changes in the anterior BNST, with Let-7a, let-7f, and miR-181a-5p upregulated in stressed female mice. This study provides evidence of distinct molecular responses to social stress in the BNST of males and females.

Targeting 2-arachidonoylglycerol signalling in the neurobiology and treatment of depression

Kennia M. Silveira et al.

Summary: 2-AG, an atypical neurotransmitter synthesized in response to stress, can modulate neurotransmission and stress response, with exposure to chronic or intense stress increasing the risk of depression. Modulating 2-AG signaling presents a potential new therapeutic target, particularly through increasing 2-AG levels or modulating CB2 receptors.

Gene-environment interactions mediate stress susceptibility and resilience through the CaMKIIβ/TARPγ-8/AMPAR pathway

Yusuke Sakai et al.

Summary: The study explores the role of CaMKII beta in the ventral hippocampus in depression susceptibility and resilience, revealing that its modulation of TARP gamma-8 phosphorylation enhances stress resilience by increasing the expression of AMPA receptor subunit GluA1. These findings provide new insights into the molecular mechanisms underlying chronic stress-induced behavioral changes and offer strategies for preventing and treating stress-related psychiatric disorders.

Summary: Research has shown a relationship between early life stress and depression, with epigenetic mechanisms playing a key role. These mechanisms involve DNA methylation, histone actions, and RNA regulation, and are of significant importance in understanding the genetic influence on psychopathology.
