Ferroptosis: molecular mechanisms and health implications

Daolin Tang et al.

Summary: Cell death can be executed through different subroutines, such as ferroptosis which is an iron-dependent form of non-apoptotic cell death. Ferroptosis can occur through two major pathways, driven by redox imbalance and abnormal expression of redox-active enzymes. The process is precisely regulated at multiple levels, with the transcription factor NFE2L2 playing a central role in anti-ferroptotic defense.

Potential therapeutic effects of Nrf2 activators on intracranial hemorrhage

Takahiko Imai et al.

Summary: Intracranial hemorrhage (ICH) is a devastating disease categorized into primary brain injury (PBI) and secondary brain injury (SBI), with oxidative and inflammatory stresses playing central roles. Nrf2 activators have the therapeutic potential in treating ICH patients by up-regulating anti-oxidative enzymes in the brain.

Iron in immune cell function and host defense

David Haschka et al.

Summary: Iron plays a crucial role in the interaction between host, pathogen, and immune responses. Restricting iron availability to invading pathogens is a powerful defense mechanism known as nutritional immunity. Iron also influences immune cell function and differentiation, as well as inflammation control.

Selective Ferroptosis Inhibitor Liproxstatin-1 Attenuates Neurological Deficits and Neuroinflammation After Subarachnoid Hemorrhage

Yang Cao et al.

Summary: Ferroptosis has been shown to contribute to neuronal damage after SAH, while liproxstatin-1 protects neurons from injury by preserving mitochondrial functions and ameliorating lipid peroxidation. Additionally, liproxstatin-1 alleviates neurological deficits and brain edema, reduces neuronal cell death, and restores redox equilibrium after SAH.

An arylthiazyne derivative is a potent inhibitor of lipid peroxidation and ferroptosis providing neuroprotection in vitro and in vivo

Meike Hedwig Keuters et al.

Summary: ADA-409-052 is a small molecule that inhibits lipid peroxidation and protects against ferroptotic cell death in neuronal cells. It also suppresses pro-inflammatory activation of microglia and reduces infarct volume and expression of pro-inflammatory genes in a mouse model of stroke. Targeting ferroptosis may be a promising therapeutic strategy for neurological diseases involving severe neuronal death and neuroinflammation.

Microglia: A Double-Edged Sword in Intracerebral Hemorrhage From Basic Mechanisms to Clinical Research

Jiachen Liu et al.

Summary: Research suggests that microglia may shift towards a pro-inflammatory phenotype following intracerebral hemorrhage, while anti-inflammatory microglia with neuroprotective effects are suppressed. Driving microglia towards an anti-inflammatory phenotype could potentially restrict inflammation and engulf cellular debris.
