A multifactorial model of pathology for age of onset heterogeneity in familial Alzheimer's disease

Diego Sepulveda-Falla et al.

Summary: PSEN1-E280A carriers exhibit a bimodal distribution of disease onset age, with no clinical differences between different age groups. Although mutant PSEN1 affects the production of Aβ, there are no significant differences in the generation and deposition of Aβ between groups. However, early-onset patients show severe hyperphosphorylated Tau pathology, while late-onset patients exhibit lesser pTau pathology and a distinct kinase activity.

The NLRP3 inflammasome drives inflammation in ischemia/reperfusion injury after transient middle cerebral artery occlusion in mice

Maximilian Franke et al.

Summary: The study demonstrates that induction of NLRP3 inflammasome in neurons plays a crucial role in neuroinflammation during acute ischemic stroke. Early blockade of NLRP3 can protect against I/R injury by reducing inflammation and stabilizing the blood-brain barrier.

Oxysterols present in Alzheimer's disease brain induce synaptotoxicity by activating astrocytes: A major role for lipocalin-2

Erica Staurenghi et al.

Summary: Among the brain hallmarks of Alzheimer's disease, reactive astrocytes are correlated with neuronal loss and cognitive deficits, while alterations in brain cholesterol metabolism contribute to AD pathogenesis. Oxysterols in AD brains induce astrocyte reactivity and the release of mediators that impact neuronal health and synapses, with lipocalin-2 playing a key role in mediating the synaptotoxic effect of oxysterol-treated astrocytes.

Prevention properties on cerebral ischemia reperfusion of medicine food homologous Dioscorea yam-derived diosgenin based on mediation of potential targets

Xinxin Zhang et al.

Summary: This study investigated the potential protective effects of diosgenin from Dioscorea yam in cerebral ischemia reperfusion injury through global proteomic analysis in a rat model. The dysregulated protein expression patterns, particularly those related to the nervous system, may provide insights for the development of strategies to prevent and treat ischemic stroke.

Ischemia-induced upregulation of autophagy preludes dysfunctional lysosomal storage and associated synaptic impairments in neurons

Xia Zhang et al.

Summary: This study revealed that in a model of oxygen-glucose deprivation, autophagy dysfunction leads to impairment of synaptic structures. Dysfunctional lysosomal storage during ischemic stress affects the balance of autophagy-lysosomal machinery crucial for maintaining neuronal synaptic function.
