Summary: Cobalamin plays a crucial role in regulating metabolism and gene expression in M. smegmatis, affecting the synthesis of methionine through the control of MetE expression. The bacterium is capable of de novo cobalamin biosynthesis during aerobic growth and limited uptake of exogenous cyanocobalamin and its precursors. The metabolic differences observed between M. smegmatis and M. tuberculosis provide valuable insights into the evolution of mycobacterial metabolism for pathogenicity.
