Resting state hypothalamic and dorsomedial prefrontal cortical connectivity of the periaqueductal gray in cocaine addiction

Summary: The study reveals that cocaine-dependent individuals experience anxiety and dysphoria during withdrawal. Compared to healthy controls, there are significant differences in the resting state functional connectivity of the periaqueductal gray, which is negatively correlated with tonic cocaine craving and recent cocaine use. Higher connections with the hypothalamus and frontoparietal regions may reflect a compensatory process to regulate craving and compulsive drug use in cocaine addiction.

Drug Addiction: Hyperkatifeia/Negative Reinforcement as a Framework for Medications Development

Summary: Compulsive drug seeking behavior associated with addiction may be driven by a heuristic framework involving three stages and three domains of dysfunction, with hyperkatifeia playing a role as a source of motivation through negative reinforcement. Neurochemical/neurocircuitry dysregulations could mediate the occurrence of a negative hedonic set point, ultimately leading to compulsive drug seeking behavior.

Summary: Using neural imaging techniques, a study explored the neural network underlying alcoholism onset in adolescents. It identified a network centered on the medial orbitofrontal cortex, with evidence of inhibitory and excitatory coregulation by this region over the dorsal periaqueductal gray. Additionally, the study found significant relationships between baseline excitatory coregulation in this network and impulsivity, supporting the role of negative urgency in alcohol dependence.
