Direct DNA crosslinking with CAP-C uncovers transcription-dependent chromatin organization at high resolution

Qiancheng You et al.

Summary: CAP-C method improves resolution and signal-to-noise in chromosome conformation capture experiments by using multifunctional DNA crosslinkers of defined sizes. It reveals high-resolution organization of chromatin compartments and shows that transcription inhibition leads to greater loss of contacts in nonloop domains. CAP-C can detect native chromatin conformation without formaldehyde prefixing.
