De novo design of modular and tunable protein biosensors

Alfredo Quijano-Rubio et al.

Summary: Protein switches have been repurposed for biosensor development by inverting the flow of information, creating modular molecular devices with a closed dark state and an open luminescent state. These sensors, based on thermodynamic coupling, require only one target binding domain for direct readout in solution, allowing for the detection of various molecules clinically, including the SARS-CoV-2 spike protein with high sensitivity and a 50-fold higher luminescence signal than background level. The modularity and sensitivity of this platform enable rapid sensor construction for a wide range of analytes, demonstrating the power of de novo protein design in creating multi-state protein systems with new functions.

Engineering luminescent biosensors for point-of-care SARS-CoV-2 antibody detection

Susanna K. Elledge et al.

Summary: A rapid and low-cost solution-based assay has been developed to detect SARS-CoV-2 antibodies in serum, plasma, whole blood, and to some extent saliva, using split luciferase antibody biosensors. This new method generates quantitative results in 30 minutes, simplifying the complexity and improving the scalability of COVID-19 antibody tests. It is suitable for point-of-care testing, population screening, and use in low-resource settings for monitoring host humoral responses to vaccination or viral infection.

Testing at scale during the COVID-19 pandemic

Tim R. Mercer et al.

Summary: The assembly and publication of the SARS-CoV-2 genome in January 2020 allowed for immediate development of tests and led to the largest global testing programme in history. Despite efforts to control the spread, widespread community transmission of the virus has become entrenched in many countries, necessitating population-scale testing to identify and isolate infected individuals. Testing plays a crucial role in the response to the COVID-19 pandemic and is likely to continue to be vital in public health.

A homogeneous split-luciferase assay for rapid and sensitive detection of anti-SARS CoV-2 antibodies

Zhong Yao et al.

Summary: A tri-part Nanoluciferase-based assay named SATiN is developed for detecting antibodies against SARS-CoV-2, showing similar sensitivity to ELISA and consistent results with various neutralizing antibody assays. This assay, performed directly in patient sera, offers rapid, quantitative, and cost-effective detection, suggesting potential applications in diagnostics and disease management.

Homogeneous Immunoassay Using a Tri-Part Split-Luciferase for Rapid Quantification of Anti-TNF Therapeutic Antibodies

Sun Jin Kim et al.

Summary: Detection of anti-TNF therapeutics is crucial in clinical treatment, but current methods are technically rigorous, slow, and expensive, limiting monitoring frequency and utility. To address this issue, researchers developed a new homogeneous immunoassay with a wide dynamic range and sufficient sensitivity for effective monitoring of serum concentrations of anti-TNF antibodies.
