Promoter-proximal CTCF binding promotes distal enhancer-dependent gene activation

Summary: CTCF, together with the cohesin complex, drives chromatin loops and topologically associating domains formation. Acute loss of CTCF affects transcription of genes that display promoter-proximal CTCF binding and are dependent on long-distance enhancers. Promoter-proximal CTCF binding is correlated with transcription of over 2,000 genes across diverse adult tissues.

Three-dimensional folding dynamics of the Xenopus tropicalis genome

Summary: The study investigates the dynamics of TADs in Xenopus tropicalis embryos and finds that TAD establishment is similar to other animals and does not rely on zygotic genome transcriptional activation. Higher self-interaction frequencies at one end of the boundary correspond to higher DNA occupancy of architectural proteins. Chromatin remodeling is essential for de novo TAD formation, and TAD structures vary in different tissues. Hi-C and single-molecule sequencing provide insights into the Xenopus tropicalis genome and three-dimensional genome dynamics during embryogenesis.
