Treatment with an inhibitor of PDGFR signaling, imatinib, reduced seizure susceptibility and suppressed microglial activation in the injured hippocampus, highlighting the potential therapeutic target for TBI-induced complications.

Summary: The study suggests that lithium may serve as a therapeutic candidate for treating blood-brain barrier breakdown in the early stage of ischemic stroke following reperfusion therapy. Lithium upregulates endothelial Wnt/beta-catenin signaling, enhances BBB integrity, reduces infarct volume, and improves neurological outcomes in mice with middle cerebral artery occlusion/reperfusion injury. Ultimately, the protective effect of lithium on BBB integrity relies on Gpr124-mediated endothelial Wnt/beta-catenin signaling.
