A randomised trial of 4-versus 12-weekly administration of bone-targeted agents in patients with bone metastases from breast or castration-resistant prostate cancer

Mark Clemons et al.

Summary: The study compared the efficacy of bone-targeted agents (BTAs) given every 4 weeks versus 12 weeks in patients with bone metastases from breast or castration-resistant prostate cancer. The results showed no significant difference in health-related quality of life, pain, global health status, rates of symptomatic skeletal events, and time to symptomatic skeletal events between the two dosing regimens. The findings suggest that de-escalation of commonly used BTAs is a viable treatment option.

Matrix stiffness-mediated effects on macrophages polarization and their LOXL2 expression

Xiaoxia Xing et al.

Summary: The study showed that increased matrix stiffness significantly enhanced M2 macrophage polarization and promoted LOXL2 expression. Activation of the integrin beta 5-FAK-MEK1/2-ERK1/2 pathway contributed to matrix stiffness-mediated HIF-1 alpha upregulation, leading to a significant improvement in LOXL2 expression.

The fibrotic and immune microenvironments as targetable drivers of metastasis

Luke Boulter et al.

Summary: Cancer cells exist within a complex microenvironment of stromal cells and extracellular matrix, where interactions between stromal cell subtypes and signaling pathways play a crucial role in controlling metastasis. The heterogeneity and plasticity within stromal cell populations add to the complexity and provide opportunities for new therapeutic approaches for targeting metastatic disease. Understanding these networks is essential for developing effective treatments.

Basement membrane stiffness determines metastases formation

Raphael Reuten et al.

Summary: Basement membrane stiffness, influenced by the ratio of netrin-4 to laminin, plays a crucial role in regulating cancer cell invasion and patient survival, surpassing the impact of pore size. Netrin-4 softens basement membranes, reducing cancer cell invasion potential despite creating larger pores, highlighting its significance in metastasis formation across various tissues.

Tumour-associated macrophages drive stromal cell-dependent collagen crosslinking and stiffening to promote breast cancer aggression

Ori Maller et al.

Summary: Stromal stiffening in malignant tumours is associated with poor treatment outcomes and increased tumour aggression. Identifying factors that regulate stromal stiffening could lead to biomarkers for patient stratification and interventions to improve outcomes. Experimental studies have shown that therapeutic targeting of tumour-associated macrophages and collagen crosslinking enzymes can reduce metastasis and stromal stiffening in aggressive breast cancer subtypes with stiff stroma. Additionally, the expression of lysyl hydroxylase 2 in the stroma has been linked to disease-specific mortality in breast cancer patients, indicating its potential as a stromal biomarker.

Integrin alpha5 in human breast cancer is a mediator of bone metastasis and a therapeutic target for the treatment of osteolytic lesions

Francesco Pantano et al.

Summary: This study identified ITGA5 as a mediator of breast-to-bone metastasis, with high expression correlating with poor bone metastasis-free survival. Silencing ITGA5 impaired tumor cell functions and bone metastasis, while inhibition with M200 showed similar effects.

Summary: The study revealed orientational ordering changes in protein structures in the metastatic environment of the lungs, which can be detected through FTIR and FTIR PCI imaging techniques. This method is useful for evaluating molecular changes in proteins that occur sequentially during the metastatic process.

Suppression of pancreatic ductal adenocarcinoma growth and metastasis by fibrillar collagens produced selectively by tumor cells

Chenxi Tian et al.

Summary: The ECM of pancreatic ductal adenocarcinoma has partially uncleaved C-terminal prodomains of fibrillar collagens, indicating reduced procollagen C-proteinase activity. The enzyme BMP1 selectively suppresses tumor growth and metastasis in cells expressing high levels of COL1A1. Cancer-cell-derived fibrillar collagen plays a role in selectively restraining tumor growth.
