Structure and mechanism of the proton-driven motor that powers type 9 secretion and gliding motility

Rory Hennell James et al.

Summary: The study reveals the internal structure and working mechanism of the gliding motility/type 9 protein secretion system motor, demonstrating that proton flow results in rotation of the GldM/PorM dimer inside the intra-membrane ring to drive processes at the bacterial outer membrane.

Site-Directed Cross-Linking Identifies the Stator-Rotor Interaction Surfaces in a Hybrid Bacterial Flagellar Motor

Hiroyuki Terashima et al.

Summary: The study directly demonstrates the physical interaction between specific residues in FliG and PomA/MotA using site-directed photo-cross-linking and disulfide cross-linking methods. The results provide insight into how stator units assemble around the rotor and generate torque in the bacterial flagellar motor.

Protein Export via the Type III Secretion System of the Bacterial Flagellum

Manuel Halte et al.

Summary: The T3SS utilizes proton motive force to export substrate proteins across the inner membrane of pathogenic bacteria. It consists of an export gate and a soluble ATPase complex in the cytoplasm. Recent research on T3SS focuses on energy transduction and protein transport across the cytoplasmic membrane.

Ton motor complexes

Anna C. Ratliff et al.

Summary: The Ton complex is a molecular motor in Gram-negative bacteria that uses the proton gradient to transport nutrients into the periplasmic space. Recent reports support a subunit stoichiometry of 5:2 for the ExbBExbD subcomplex. This review summarizes recent discoveries of the structures and mechanisms of the Ton system, as well as similar protein motor complexes in Gram-negative bacteria.

Mechanosensitive remodeling of the bacterial flagellar motor is independent of direction of rotation

Navish Wadhwa et al.

Summary: Motility is crucial for the survival and dispersal of many bacteria, and plays a role during infections. Recent research has shown that the bidirectional flagellar motor of E. coli adjusts torque-generating units in response to changes in load, representing a robust strategy for quickly regulating output.

Molecular structure of the intact bacterial flagellar basal body

Steven Johnson et al.

Summary: The intact Salmonella flagellar basal body structure, including the inner membrane rotor, drive shaft, and outer membrane bushing complex, has been elucidated by in situ cryo-electron microscopy. This complex macromolecular structure enables bacterial motility by assembling a strong, multicomponent drive shaft that powers bacterial swimming in viscous fluids. The molecular details of how different protein molecules assemble into a complex structure spanning two membranes and a cell wall have been revealed.
