Myeloma-Modified Adipocytes Exhibit Metabolic Dysfunction and a Senescence-Associated Secretory Phenotype

Heather Fairfield et al.

Summary: Interactions between bone marrow adipocytes (BMAds) and myeloma cells play a significant role in the pathogenesis and treatment of multiple myeloma. Studies show that myeloma cells reduce bone marrow adipose tissue (BMAT) and induce senescence and metabolic changes in BMAds.

Maturation conditions, post-ovulatory age, medium pH, and ER stress affect [Ca2+]i oscillation patterns in mouse oocytes

Rui-Ying Yuan et al.

Summary: The study found that oocytes released to the oviduct at 17 hours post-hCG showed stronger [Ca2+]i oscillation. Oscillations in acidic conditions were weaker, while in vitro-matured oocytes had lower frequency and peak. Endoplasmic reticulum stress severely affected [Ca2+]i oscillation parameters.

Emodin-Induced Oxidative Inhibition of Mitochondrial Function Assists BiP/IRE1α/CHOP Signaling-Mediated ER-Related Apoptosis

Li-zhen Qiu et al.

Summary: The study found that emodin-induced oxidative stress activated ER stress and the UPR through the BiP/IRE1 alpha/CHOP signaling pathway, resulting in ER Ca2+ release and cytoplasmic Ca2+ overloading. Additionally, redox imbalance caused by emodin increased mtROS while decreasing function, leading to leakage of proapoptotic factors. Blocking Ca2+ release from ER could inhibit emodin-induced apoptosis, indicating potential targets for treatment of liver injury induced by Cassia seeds and related substances.

Endoplasmic reticulum stress signals in the tumour and its microenvironment

Xi Chen et al.

Summary: This review explores how ER stress in the tumor microenvironment can influence the functions of cancer cells and immune cells, leading to vulnerabilities that could be targeted by emerging therapeutic strategies. The tightly regulated processes of protein handling, modification, and folding in the ER determine cell function and survival, and disruptions in ER homeostasis can result in a state of persistent ER stress with pro-tumoral effects in cancer cells and immunomodulatory effects in immune cells.
