The toxicity of cell therapy: Mechanism, manifestations, and challenges

Yongjia Jin et al.

Summary: Adoptive cell therapy (ACT) has shown significant clinical benefits for cancer treatment, but is often associated with toxicities ranging from mild to life-threatening. Common toxicities include cytokine release syndrome (CRS), neurological toxicity, and on-target or off-target toxicities. This review summarizes the clinical manifestations and underlying pathological mechanisms of these adverse events, and discusses the challenges and opportunities of managing ACT-related toxicities.

Summary: The study shows that TKIs have a negative impact on oocyte and sperm maturation, gonadal function, and overall fertility potential, with a lower risk of teratogenicity to a fathered pregnancy with TKI use. The complexity of pregnancy outcomes and results in women taking TKIs compared to men is higher, and it is advised for women to avoid conception while taking TKIs.
