Engineered Nanoparticle Applications for Recombinant Influenza Vaccines

Zachary R. Sia et al.

Summary: The combination of recombinant biotechnology and nanomedicine techniques shows promise in developing a next-generation flu vaccine with broad protection. Various nanoparticle candidates have been tested, showing protection against different influenza strains, production of antibodies with broad reactivity, and activation of potent T cell responses. The future of influenza vaccines may involve combining recombinant antigens with nanoparticle carriers.

Experimental and Computational Observations of Immunogenic Cobalt Porphyrin Lipid Bilayers: Nanodomain-Enhanced Antigen Association

Jasmin Federizon et al.

Summary: CoPoP facilitates non-covalent surface-display of antigens on liposomes, enhancing vaccine efficacy. Compared to aquocobalamin or cobalt nitrilotriacetic acid (Co-NTA) liposomes, Pfs25 binds more strongly to CoPoP liposomes, enhancing antigen internalization by macrophages. Molecular dynamics simulations reveal that CoPoP-containing bilayers create nanodomains allowing for efficient stabilization of antigens.

Hemagglutinin Functionalized Liposomal Vaccines Enhance Germinal Center and Follicular Helper T Cell Immunity

Mai N. Vu et al.

Summary: This study presents a facile method to produce lipid nanoparticle subunit vaccines that exhibit high immunogenicity and provide protection against influenza virus. The vaccines showed increased serum antibody titers and superior protection against highly pathogenic viruses, along with enhanced B cell and follicular helper T cell responses. This work facilitates the rational design and rapid production of next generation nanoparticle subunit vaccines.

Quadrivalent influenza nanoparticle vaccines induce broad protection

Seyhan Boyoglu-Barnum et al.

Summary: Computational design of nanoparticle immunogens can induce potent and broad antibody responses against diverse influenza viruses, including neutralizing antibody responses to vaccine-matched strains and broadly protective antibody responses to heterologous viruses. These novel vaccines have the potential to replace traditional seasonal vaccines.
