Summary: Many studies have shown that negative allosteric modulators (NAM) of metabotropic glutamate receptor 5 (mGlu5) can reduce cocaine and methamphetamine seeking in extinction-reinstatement animal models of addiction. The study investigated the role of mGlu5 in the incubation of methamphetamine craving during abstinence, revealing differences from cocaine craving incubation. Systemic administration of mGlu5 NAM reduced incubated methamphetamine seeking, but also reduced inactive behaviors and locomotion, while intra-NAc core administration had no significant effects.
