Antiviral activity of amides and carboxamides of quinolizidine alkaloid (-)-cytisine against human influenza virus A (H1N1) and parainfluenza virus type 3

Summary: Novel derivatives of quinolizidine alkaloid (-)-cytisine were synthesised and evaluated for their activity against influenza virus and human parainfluenza virus. Compounds with phenyl and allyl urea's fragments showed the highest activity against influenza virus, possibly due to their predicted interactions with the active site of the virus enzyme.

Summary: The alpha 7 and alpha 4 beta 2* nicotinic acetylcholine receptors are the most abundant nAChRs in the mammalian brain and are targeted in drug discovery for brain disorders. A C(10) variant of cytisine has been developed as a partial agonist of alpha 4 beta 2 nAChR. Molecular dynamic simulations and experiments revealed the structural basis for the selectivity of 10-methylcytisine for alpha 7 and alpha 4 beta 2 nAChRs, providing insights into agonist binding and receptor dynamics.

Summary: Natural products and their derivatives are important in discovering new biologically active compounds, with cytisine being synthesized for new drug development. QSAR modeling was used to predict and select active cytisine-containing 1,3-oxazoles, with promising results. Compounds 10, 11 showed high activity as potential inhibitors of Candida spp., demonstrating potential as antifungal agents.
