Validated RP-HPLC Method Development for Gentamicin Microanalysis in Pharmaceutical Dosage Forms, Urine and Blood

An accurate and reliable HPLC method was developed for quantifying gentamicin in biological fluids and pharmaceuticals. The method showed high sensitivity and excellent recovery rates for the spiked drug.
Gentamicin is an important member of the aminoglycoside class of antibiotics having broader spectrum with extend to ototoxicity and nephrotoxcity. A reliable and accurate high performance liquid chromatography (HPLC) procedure was established using Agilent Zorbax Eclipse plus phenyl hexyl C18 (4.6 x 250 mm, 5 mu m) column and UV detection at 330 nm. Recovery of gentamicin from biological and pharmaceutical samples was more than 95-98% after spiking the drug. The sensitivity of method is predicted by calculating a lower level of the limit of detection and lower limit of quantification (0.286 and 0.867 mu g/mL, respectively. The procedure was used to quantify gentamicin in biological fluids and pharmaceuticals dosage form.
