Beyond small molecules: targeting G-quadruplex structures with oligonucleotides and their analogues

Cognate base-pair selectivity of hydrophobic unnatural bases in DNA ligation by T4 DNA ligase

Summary: Our study demonstrates the cognate base pair selectivity of T4 DNA ligase using a hydrophobic unnatural base pair (Ds-Pa) for template-dependent ligation. The efficient recognition of Ds-Pa pairing and exclusion of noncognate pairings by Ds indicate higher selectivity compared to natural base pairings. This hydrophobic base pairing can be utilized in enzymatic ligation for genetic alphabet expansion, paving the way for the creation of semi-synthetic organisms containing unnatural base pairs.
