AAV delivery of shRNA against IRS1 in GABAergic neurons in rat hippocampus impairs spatial memory in females and male rats

Sandra Sanchez-Sarasua et al.

Summary: Brain insulin resistance is closely related to impaired cognitive function and the onset of Alzheimer's disease. The specific effect of impaired IRS1 function in an otherwise healthy brain has not been fully understood. Studies have shown that insulin resistance is associated with inflammation, and even small alterations in the insulin/IGF1 pathway in specific hippocampal circuitries may lead to changes in synaptic plasticity and behavior.

Tauroursodeoxycholic Acid Attenuates Diet-Induced and Age-Related Peripheral Endoplasmic Reticulum Stress and Cerebral Amyloid Pathology in a Mouse Model of Alzheimer's Disease

Summary: Interventions targeting peripheral ER stress can alleviate the accumulation of Aβ in the brain induced by obesity and diabetes, as well as effectively reduce age-related obesity and insulin resistance, significantly reducing amyloid accumulation.
