A dysfunctional TRPV4-GSK3β pathway prevents osteoarthritic chondrocytes from sensing changes in extracellular matrix viscoelasticity

Pranay Agarwal et al.

Summary: The viscoelasticity of alginate hydrogels regulates cell volume in healthy chondrocytes but not in osteoarthritic chondrocytes. This difference in response is associated with changes in anabolic gene expression, secretion of pro-inflammatory cytokines, and modulation of intracellular calcium. Dysfunctional TRPV4-GSK3 beta pathway in osteoarthritic chondrocytes renders them unable to respond to viscoelastic changes in the extracellular matrix.

Mechanosensory and mechanotransductive processes mediated by ion channels in articular chondrocytes: Potential therapeutic targets for osteoarthritis

Kun Zhang et al.

Summary: Articular cartilage is composed of an extracellular matrix with embedded chondrocytes, and its formation and function are influenced by mechanical forces. Hind limb unloading or simulated microgravity can lead to cartilage loss, emphasizing the importance of a healthy mechanical environment and appropriate mechanical stimulation for maintaining cartilage homeostasis. Mechanosensitive ion channels play a significant role in regulating the metabolism of articular chondrocytes and can be influenced by various mechanical stimuli, ultimately impacting the progression of osteoarthritis.
