Plasma IL-12/IFN-γ axis predicts cognitive trajectories in cognitively unimpaired older adults

Hyun-Sik Yang et al.

Summary: The study found that IL-12p70 and IFN-γ are associated with cognitive decline in cognitively unimpaired elderly, and this association may be related to Aβ levels. This suggests that immune dysregulation plays an important role in early-stage cognitive decline.

Presenilin 1 phosphorylation regulates amyloid-β degradation by microglia

Jose Henrique Ledo et al.

Summary: Microglia containing phospho-deficient mutant PS1 show a slower kinetic response to brain micro injury in vivo and an inability to degrade A beta oligomers, leading to severe A beta accumulation in microglia in an Alzheimer's mouse model. This demonstrates a novel mechanism by which PS1 modulates microglial function and contributes to Alzheimer's-associated phenotypes.

Dissecting Alzheimer's disease pathogenesis in human 2D and 3D models

Giovanna Cenini et al.

Summary: Alzheimer's disease is on the rise with the aging population, posing as a major health concern in modern society. The use of human pluripotent stem cells and cell reprogramming offers new prospects for understanding the pathogenic mechanisms of the disease and developing effective therapies. Advances in 3D cell culture systems, including organoids and neurospheroids, are overcoming limitations in modeling extracellular disease-related alterations, providing new opportunities for disease research and therapy development.

Increased serum peripheral C-reactive protein is associated with reduced brain barriers permeability of TSPO radioligands in healthy volunteers and depressed patients: implications for inflammation and depression

Summary: The studies suggest that peripheral inflammation may lead to a reduction in blood-brain barrier permeability, disrupting brain homeostasis and inducing depressive symptoms.

Hospital-treated infectious diseases and the risk of dementia: a large, multicohort, observational study with a replication cohort

Pyry N. Sipila et al.

Summary: Infections requiring hospital treatment have been found to increase the long-term risk of dementia, including vascular dementia and Alzheimer's disease, with no specificity in infection type. The association is driven by general inflammation rather than specific microbes, and there is evidence of dose-response relationships between infectious disease burden and dementia risk.

Astrocytic interleukin-3 programs microglia and limits Alzheimer's disease

Cameron S. McAlpine et al.

Summary: Communication within the glial cell ecosystem is essential for neuronal and brain health. This study demonstrates that astrocyte-sourced interleukin-3 programs microglia to ameliorate the pathology of Alzheimer's disease by enhancing their ability to clear aggregates of beta-amyloid and tau, ultimately restricting disease progression and cognitive decline. These findings highlight IL-3 as a key mediator of astrocyte-microglia cross-talk and a potential therapeutic target for Alzheimer's disease.

Gut microbiome pattern reflects healthy ageing and predicts survival in humans

Tomasz Wilmanski et al.

Summary: The study finds that as individuals age, the gut microbiome becomes increasingly unique, with this uniqueness strongly associated with microbially produced amino acid derivatives in the bloodstream. Healthy older individuals continue to show microbial drift towards a unique compositional state, while less healthy individuals do not. Retaining high levels of Bacteroides dominance or having a low gut microbiome uniqueness measure predicts decreased survival in a 4-year follow-up.
