Evaluating totipotency using criteria of increasing stringency

Summary: The article describes the criteria for defining totipotency and evaluates candidate totipotent stem cell types using different levels of stringency. It challenges the idea that expanded or extended pluripotent states have increased totipotent potential compared to conventional embryonic stem cells under in vitro and in vivo conditions.

Summary: This study found that protein synthesis rates decrease during HSC ontogeny, while erythroid protein synthesis rates increase. A ribosomal mutation impacting ribosome biogenesis can disrupt HSC self-renewal, particularly in fetal erythropoiesis. Developmental changes in protein synthesis cause hematopoietic stem and progenitor cells to have differential sensitivity to impaired ribosome biogenesis.

Spatiotemporal dissection of the cell cycle with single-cell proteogenomics

Summary: The study comprehensively mapped spatial and temporal variations among individual human cell proteomes across the cell cycle, revealing that approximately one-fifth of the human proteome displays cell-to-cell variability. The researchers identified hundreds of proteins with previously unknown associations with mitosis and the cell cycle, and found evidence suggesting that some of these proteins have oncogenic functions.
