Normal and defective pathways in biogenesis and maintenance of the insulin storage pool

Ming Liu et al.

Summary: Both basal and glucose-stimulated insulin release primarily occur through insulin secretory granule exocytosis from pancreatic beta cells, which are essential for maintaining normal blood glucose levels. Loss of insulin-secreting beta cells may result from exhaustion of insulin reserves or beta cell dedifferentiation, affecting glycemic control. Defects in biosynthetic generation and maintenance of the total insulin granule pool can also contribute to diabetes risk.

Saturated fatty acids entrap PDX1 in stress granules and impede islet beta cell function

Mu Zhang et al.

Summary: This study revealed that saturated fatty acids disrupt nucleocytoplasmic transport and induce stress granule (SG) formation in pancreatic beta cells. The formation of SGs is mediated by the phosphoinositide 3-kinase (PI3K)/eukaryotic translation initiation factor 2 alpha (EIF2 alpha) pathway, and may be linked to beta cell dysfunction. Disruption of SG assembly may be a potential therapeutic strategy for treating obesity and type 2 diabetes.

Temporal Associations Among Body Mass Index, Fasting Insulin, and Systemic Inflammation A Systematic Review and Meta-analysis

Natasha Wiebe et al.

Summary: Through analyzing 60 studies, it was found that most participants in the studies experienced decreases in BMI, fasting insulin level, and C-reactive protein level. The study suggests that changes in fasting insulin levels precede changes in weight, and there is a positive correlation between fasting insulin level and BMI, rather than obesity causing noncommunicable chronic diseases and premature death.

A New Hypothesis for Type 1 Diabetes Risk: The At-Risk Allele at rs3842753 Associates With Increased Beta-Cell INS Messenger RNA in a Meta-Analysis of Single-Cell RNA-Sequencing Data

Summary: The study found that beta cells with the rs3842753 C allele had higher INS expression and showed increased endoplasmic reticulum stress gene expression, suggesting that the inherited risk of type 1 diabetes may be associated with elevated insulin production.
