Promoter-proximal CTCF binding promotes distal enhancer-dependent gene activation

Naoki Kubo et al.

Summary: CTCF, together with the cohesin complex, drives chromatin loops and topologically associating domains formation. Acute loss of CTCF affects transcription of genes that display promoter-proximal CTCF binding and are dependent on long-distance enhancers. Promoter-proximal CTCF binding is correlated with transcription of over 2,000 genes across diverse adult tissues.

HiC-ACT: improved detection of chromatin interactions from Hi-C data via aggregated Cauchy test

Taylor M. Lagler et al.

Summary: The study introduces a novel approach called HiC-ACT that aims to improve the detection of chromatin interactions by utilizing information from neighboring chromatin interactions. By post-processing results from methods assuming independence, HiC-ACT enhances sensitivity, controls type I error, and is computationally efficient. The approach shows advantages in detecting interactions with lower signal-to-noise ratio and similar epigenetic signatures, and exhibits higher correlation with known enhancers compared to existing methods Fit-Hi-C/FitHiC2.
