Revving the CAR-Combination strategies to enhance CAR T cell effectiveness

Rajat Bansal et al.

Summary: CAR T cell therapy has shown impressive response rates in refractory B-cell malignancies, but faces challenges in solid malignancies. Experimental strategies combining small molecules and monoclonal antibodies are being explored to overcome resistance mechanisms to CAR T cells.

Counteracting CAR T cell dysfunction

Mansour Poorebrahim et al.

Summary: Despite the high rates of complete remission achieved through CAR T cell therapy, the efficacy of this approach is limited by dysfunctional CAR T cells, which can be improved through modifications to enhance antitumor functions and lifespan. Research is ongoing on blocking exhaustion and senescence in CAR T cells to boost potency in adoptive cell transfer therapies.

Low-dose decitabine priming endows CAR T cells with enhanced and persistent antitumour potential via epigenetic reprogramming

Summary: The study demonstrates that pre-treatment of CAR T cells with a DNA methyltransferase inhibitor enhances anti-tumor activities, cytokine production, and proliferation, allowing for effective eradication of tumors at low doses and improved response to tumor rechallenge.

Class I histone deacetylase inhibition promotes CD8 T cell activation in ovarian cancer

Tyler R. McCaw et al.

Summary: The study found that treatment with the class 1 HDAC inhibitor entinostat can enhance the expression of pathways and genes associated with CD8 T cell cytotoxic function, while downregulating myeloid derived suppressor cell chemoattractants. The suppressive capacity of regulatory T cells within tumors and associated ascites was significantly reduced, reversing the CD8-Treg ratio.

Bcl-2 Enhances Chimeric Antigen Receptor T Cell Persistence by Reducing Activation-Induced Apoptosis

Haiyong Wang et al.

Summary: Chimeric antigen receptor-modified T cells (CAR-T) have shown success in treating B-cell leukemia, but struggle with efficacy in lymphoma and solid tumors. This study aimed to evaluate the potential of adding anti-apoptotic molecules, like Bcl-2, to enhance CAR-T cell anti-tumor activity. Integrating Bcl-2 improved CAR-T cell proliferation, leading to enhanced anti-tumor effects in a mouse model of lymphoma.

Manipulating the Metabolism to Improve the Efficacy of CAR T-Cell Immunotherapy

Marsha Pellegrino et al.

Summary: The adoptive transfer of CAR-expressing T cells has shown unprecedented success in treating B-cell malignancies, but its efficacy in other types of cancers is still limited. The metabolic fitness of CAR T cells plays a crucial role in their ability to target and function within solid tumors, with both tumor and T cell metabolism impacting the immune response significantly.

Cell-programmed nutrient partitioning in the tumour microenvironment

Bradley Reinfeld et al.

Summary: The study found that in the tumor microenvironment, different cell types have varying abilities to access and consume glucose and glutamine, with nutrient partitioning being programmed in a cell-intrinsic manner through mTORC1 signaling and the expression of genes related to glucose and glutamine metabolism.

NAD+ metabolism, stemness, the immune response, and cancer

Lola E. Navas et al.

Summary: NAD(+) plays important roles in redox metabolism, aging, longevity, immune system, and DNA repair. Dysregulation of NAD(+) levels is associated with metabolic diseases and aging-related diseases. NAMPT is the key enzyme recovering NAD from nicotinamide. The roles of NAD(+) and NAMPT in cancer metabolism, immune system, stemness, aging, and cancer are significant.
