Chondroitinase and Antidepressants Promote Plasticity by Releasing TRKB from Dephosphorylating Control of PTPσ in Parvalbumin Neurons

Summary: Perineuronal nets (PNNs) restrict cortical network plasticity by encasing PV+ interneurons, but removal of PNNs using chondroitinase ABC (chABC) in the adult visual cortex requires intact TRKB signaling in PV+ neurons. Additionally, the interaction between protein tyrosine phosphatase sigma (PTP sigma) and TRKB limits TRKB phosphorylation, while the antidepressant drug fluoxetine disrupts this interaction to promote TRKB signaling and reopen critical period-like plasticity. By inhibiting TRKB dephosphorylation by the PTP sigma-CSPG complex, both chABC and fluoxetine facilitate critical period-like plasticity in the adult visual cortex.
