Salusin-α mitigates diabetic nephropathy via inhibition of the Akt/mTORC1/p70S6K signaling pathway in diabetic rats

Wen-Juan Wang et al.

Summary: The present study investigated the potential roles of salusin-alpha in diabetic renal disease. The levels of salusin-alpha were downregulated in plasma and kidney tissues from diabetic rats. Exogenous administration of salusin-alpha alleviated the characteristics of diabetic nephropathy, reduced renal fibrosis, suppressed reactive oxygen species (ROS) production, and counteracted the activated Akt/mTORC1/p70S6K signaling pathway. These findings suggest that salusin-alpha may have a protective effect against diabetic nephropathy.

Ferroptosis: machinery and regulation

Xin Chen et al.

Summary: Ferroptosis is an iron-dependent, non-apoptotic form of regulated cell death caused by lipid peroxidation. Lipoxygenase is the main promoter of ferroptosis, while GPX4 is the central repressor of this process.

Ginkgetin derived from Ginkgo biloba leaves enhances the therapeutic effect of cisplatin via ferroptosis-mediated disruption of the Nrf2/HO-1 axis in EGFR wild-type non-small-cell lung cancer

Jian-Shu Lou et al.

Summary: The study found that ginkgetin derived from Ginkgo biloba leaves enhances DDP-induced cytotoxicity in NSCLC cells through inducing ferroptosis. The results indicate that, in addition to enhancing DDP-induced cytotoxicity, ginkgetin can disrupt redox homeostasis in DDP-treated cells and enhance apoptosis.

Ferroptosis: molecular mechanisms and health implications

Daolin Tang et al.

Summary: Cell death can be executed through different subroutines, such as ferroptosis which is an iron-dependent form of non-apoptotic cell death. Ferroptosis can occur through two major pathways, driven by redox imbalance and abnormal expression of redox-active enzymes. The process is precisely regulated at multiple levels, with the transcription factor NFE2L2 playing a central role in anti-ferroptotic defense.

Incretin drugs in diabetic kidney disease: biological mechanisms and clinical evidence

Radica Z. Alicic et al.

Summary: Incretin-based therapies, such as GLP1R agonists and DPP4 inhibitors, have shown protective effects in diabetic kidney disease patients, with potential mechanisms including immunomodulation and anti-inflammatory actions. These drugs not only lower blood glucose levels effectively but also reduce the risk of developing chronic kidney disease and cardiovascular events, in addition to potentially attenuating oxidative stress and cellular apoptosis in the kidney.

Polysulfide-mediated sulfhydration of SIRT1 prevents diabetic nephropathy by suppressing phosphorylation and acetylation of p65 NF-κB and STAT3

Hai-Jian Sun et al.

Summary: Sodium tetrasulfide (Na2S4) treatment can ameliorate diabetic kidney disease by inhibiting inflammation cytokines and reducing oxidative stress, renal fibrosis, and cell apoptosis. Na2S4 acts through sulfhydrating SIRT1 to deactivate p65 NF-kappa B and STAT3 phosphorylation/acetylation, ultimately reducing oxidative stress, inflammation response, and EMT progression in renal tubular epithelial cells.

Inhibition of ferroptosis by up-regulating Nrf2 delayed the progression of diabetic nephropathy

Shuangwen Li et al.

Summary: Our research demonstrated the involvement of ferroptosis in the development of DN, and up-regulating Nrf2 with fenofibrate treatment inhibited diabetes-related ferroptosis, delaying the progression of DN. This study provided a new perspective on the development mechanism of DN and offered a new approach to delay the progression of DN.
