MAPK1/3 kinase-dependent ULK1 degradation attenuates mitophagy and promotes breast cancer bone metastasis

Summary: ULK1 deficiency impairs mitophagy in breast cancer cells under hypoxia, leading to accumulation of damaged mitochondria, NLRP3 inflammasome activation, and bone metastasis. Phosphorylation of ULK1 triggers its degradation, and a negative correlation between ULK1 expression levels and p-MAPK1/3 was observed. The use of MAP2K/MEK inhibitor trametinib can restore mitophagy function and inhibit NLRP3 inflammasome activation to reduce bone metastasis.
