How Pathogens Feel and Overcome Magnesium Limitation When in Host Tissues

Anne-Beatrice Blanc-Potard et al.

Summary: Host organisms use nutritional immunity to restrict the availability of essential nutrients to invading pathogens, with magnesium deprivation playing a significant role in limiting the growth of Salmonella enterica serovar Typhimurium in host tissues. Pathogens' responses to factors promoting nutritional immunity may be more reflective of their internal cytoplasmic environment rather than the nutrient concentration in host cell compartments.

Acetylation of PhoP K88 Is Involved in Regulating Salmonella Virulence

Jianhui Li et al.

Summary: The study showed that acetylation regulates the activity of PhoP in Salmonella enterica serovar Typhimurium, impacting virulence, with a conserved lysine residue, K88, playing a crucial role in this process by inhibiting PhoP dimerization and DNA-binding abilities. Additionally, mutation of K88 to mimic the acetylated form significantly reduced intestinal inflammation and systemic infection in a mouse model, highlighting the close link between virulence and metabolism in the host.

Reduced ATP-dependent proteolysis of functional proteins during nutrient limitation speeds the return of microbes to a growth state

Jinki Yeom et al.

Summary: In times of nutrient scarcity, microorganisms like Salmonella enterica serovar Typhimurium can preserve functional proteins by inhibiting their degradation, allowing for a rapid return to growth once nutrients become available. This strategy is conserved across different microbial species and facilitates quick recovery from slow-growth states.

The PhoPQ Two-Component System Is the Major Regulator of Cell Surface Properties, Stress Responses and Plant-Derived Substrate Utilisation During Development of Pectobacterium versatile-Host Plant Pathosystems

Uljana Kravchenko et al.

Summary: Pectobacterium versatile, previously known as P. carotovorum, is a versatile soft rot enterobacteria species with complex transcriptional regulation of virulence properties, mainly controlled by the PhoPQ two-component system. PhoP was found to regulate at least 115 genes specific for pectobacteria, involved in plant-derived carbon source metabolism, bacterial cell envelope modification, stress resistance, and establishment of plant cell wall decomposition order. The PhoP binding site motif was defined, showing universality in enteric bacteria. PhoP plays a crucial role as a global virulence regulator in P. versatile-host plant pathosystem.

Biphasic regulation of transcriptional surge generated by the gene feedback loop in a two-component system

Wen Liu et al.

Summary: Researchers used PhoP/PhoQ TCS as a model system and identified two signaling pathways that modulate the level of P-PhoP, with competition between pathways determining the generation of P-PhoP surge. Additionally, PhoP was found to be a bifunctional modulator that contributes to dynamic control of P-PhoP state.

Limitation of phosphate assimilation maintains cytoplasmic magnesium homeostasis

Roberto E. Bruna et al.

Summary: Phosphorus is essential for biological molecules in bacteria, but excessive intake can lead to cytoplasmic Mg2+ deficiency, affecting bacterial growth and viability. Bacteria limit Pi uptake to prevent this, showing a regulatory logic based on the connection to cytoplasmic Mg2+ homeostasis.

MAPS integrates regulation of actin-targeting effector SteC into the virulence control network of Salmonella small RNA PinT

Sara Correia Santos et al.

Summary: The study reveals the important role of the Hfq-associated sRNA PinT in Salmonella virulence, showing its translational inhibition of the secreted effector kinase SteC and delay of actin rearrangements in infected host cells. The findings highlight the need for complementary methods to uncover the full target suites of sRNAs.

Low Cytoplasmic Magnesium Increases the Specificity of the Lon and ClpAP Proteases

Jinki Yeom et al.

Summary: In Salmonella enterica serovar Typhimurium, the regulation of proteolysis specificity by the HspQ protein and the PhoP regulator in response to low cytoplasmic Mg2+ plays a critical role in controlling protein synthesis and antibiotic persistence.PhoP is a transcriptional regulator that governs proteostasis, and its activation in low cytoplasmic Mg2+ condition impacts the abundance of preexisting proteins by affecting the Lon and ClpAP proteases and their substrates. This mechanism is conserved in enteric bacteria and provides insights into bacterial adaptation to changing environmental conditions.

Effects of Regulatory Network Organization and Environment on PmrD Connector Activity and Polymyxin Resistance in Klebsiella pneumoniae and Escherichia coli

Annie Chen et al.

Summary: This study explores the role of PmrD in modulating polymyxin resistance in Klebsiella pneumoniae and Escherichia coli. In K. pneumoniae, PmrD is not necessary for polymyxin resistance due to mgrB mutations, but contributes under conditions of low pH or intermediate iron concentrations. In E. coli, PmrD acts as a connector between PhoQ/PhoP and PmrB/PmrA, with activity depending on PmrB/PmrA stimulation or high activation of PhoQ/PhoP. These findings suggest that the role of the PmrD connector in modulating the polymyxin resistance network varies depending on the organism and environmental conditions associated with PmrB stimulation.
