SARS-CoV-2 S1 is superior to the RBD as a COVID-19 subunit vaccine antigen

Yunfei Wang et al.

Summary: The S1 domain of SARS-CoV-2 induces higher levels of antibodies in mice and efficiently neutralizes the virus when adjuvanted with alum. Research suggests that expression systems with posttranslational modification abilities and adjuvants prone to a Th1 response are crucial for effective COVID-19 vaccines.

SARS-CoV-2 variants with mutations at the S1/S2 cleavage site are generated in vitro during propagation in TMPRSS2-deficient cells

Michihito Sasaki et al.

Summary: This study identified SARS-CoV-2 variants with mutations at the cleavage motif of the S protein (S gene mutants) that undergo inefficient proteolytic cleavage, leading to smaller plaques and less infectivity. These mutants emerged rapidly through virus propagation in Vero cells and may have been unknowingly used in experiments, antiviral screenings, and vaccine development. The mutations affect the virus' entry pathway and cell tropism, emphasizing the importance of monitoring de novo S gene mutations during tissue-culture propagation.

Mink, SARS-CoV-2, and the Human-Animal Interface

Florence Fenollar et al.

Summary: Mink, small carnivores of the Mustelidae family, have been widely bred for their fur. Outbreaks of SARS-CoV-2 have been reported in mink farms in multiple countries, raising concerns about their potential role in the pandemic and the impact of mutant viruses on various aspects of the disease.

Protection of K18-hACE2 mice and ferrets against SARS-CoV-2 challenge by a single-dose mucosal immunization with a parainfluenza virus 5-based COVID-19 vaccine

Dong An et al.

Summary: This mucosal vaccine strategy inhibits SARS-CoV-2 replication in the upper respiratory tract, preventing disease transmission. The vaccine candidate CVXGA1 shows promising protective effects in animal models.

A bivalent recombinant vaccine targeting the S1 protein induces neutralizing antibodies against both SARS-CoV-2 variants and wild-type of the virus

Cai He et al.

Summary: The study found that the S1-Mut antigens were more effective in inhibiting ACE2 binding in the B.1.351 variant. Additionally, the bivalent vaccine exhibited ideal neutralization properties against the wild-type and B.1.351 variant, as well as other variants.

SARS-CoV-2 Glycosylation Suggests That Vaccines Should Have Adopted the S1 Subunit as Antigen

Ariel Fernandez

Summary: Current SARS-CoV-2 vaccines use the trimeric spike protein as antigen, but due to glycosylation and epitope diversity, the immune response is somewhat limited. The receptor binding domain within the S1 subunit is the most exposed to antibody recognition, offering more potential epitopes for antibody exposure post-virus activation. Vaccines adopting the S1 subunit as antigen may be more efficacious in generating a strong immune response.
