The effect of IFN-β 1a on expression of MDA5 and RIG-1 in multiple sclerosis patients

Gholamreza Asadikaram et al.

Summary: The study compared mRNA levels of MDA5 and RIG-1 in MS patients and healthy controls, finding decreased levels in patients. IFN-beta 1a therapy led to upregulation of RIG-1, especially in female patients, suggesting a protective effect against infectious diseases.

Regulation of RIG-I-like receptor-mediated signaling: interaction between host and viral factors

Koji Onomoto et al.

Summary: RLRs are RNA sensor molecules that play essential roles in innate antiviral immunity by inducing downstream signaling via interactions with various host and viral factors. The signaling mediated by RLRs is regulated by interactions with endogenous RNAs and host proteins, and deregulation can lead to autoimmune and autoinflammatory disorders.

Viral Evasion of RIG-I-Like Receptor-Mediated Immunity through Dysregulation of Ubiquitination and ISGylation

Cindy Chiang et al.

Summary: Viral manipulation of ubiquitin or ISG15-mediated immune responses is a key strategy for virus evasion, including direct targeting of ubiquitin E3 ligases, active deubiquitination using viral DUBs, and the upregulation of cellular DUBs to regulate RLR signaling. Certain coronaviruses, such as SARS-CoV-2, actively deISGylate key molecules in the RLR pathway to escape type I interferon-mediated antiviral responses. Targeting virally-encoded proteins that manipulate ubiquitin or ISG15-mediated innate immune responses may be a potential strategy for the development of new antivirals and vaccines.

ISG15-dependent activation of the sensor MDA5 is antagonized by the SARS-CoV-2 papain-like protease to evade host innate immunity

GuanQun Liu et al.

Summary: Conjugation of ISG15 plays a crucial role in activating the RIG-I-like receptor MDA5 and triggering antiviral responses, while SARS-CoV-2 suppresses MDA5 activation through direct de-ISGylation to evade innate immunity.

SARS-CoV-2 membrane glycoprotein M antagonizes the MAVS-mediated innate antiviral response

Yu-Zhi Fu et al.

Summary: A novel SARS-related coronavirus (SARS-CoV-2) has emerged as a serious pathogen causing high morbidity and mortality. The membrane glycoprotein M of SARS-CoV-2 was identified as a negative regulator of the innate immune response by impairing MAVS aggregation and downstream signaling, thus evading the innate antiviral response.
