Sheathless CE-MS as a tool for monitoring exchange efficiency and stability of bispecific antibodies

Summary: The study explored the potential of sheathless CE-MS for monitoring exchange efficiency and stability of in-house produced bispecific antibodies. The method successfully separated BsAbs from their parent monospecific antibodies using a polyethyleniimine (PEI)-coated capillary and acidic background electrolytes, allowing reliable assessment of the exchange efficiency and monitoring of stability under different storage conditions. The method was especially valuable for a Fab-glycosylated BsAb due to difficulties in discriminating between the highly heterogeneous glycans and monospecific parent antibody masses.

Summary: Urinary peptides have gained attention as potential biomarkers in kidney and cardiovascular disease, with capillary electrophoresis coupled mass spectrometry being the most commonly used technology. The most abundant urinary peptides are typically collagen fragments, reflecting inflammation and fibrosis. The successful completion of the first urinary peptide guided intervention trial, PRIORITY, is expected to further drive clinical application for early detection of chronic diseases, progression prediction, and drug response prognosis.

Use of Capillary Zone Electrophoresis Coupled to Electrospray Mass Spectrometry for the Detection and Absolute Quantitation of Peptidoglycan-Derived Peptides in Bacterial Cytoplasmic Extracts

Summary: Peptidoglycan (PGN) is a crucial structure in bacterial cell walls, continuously synthesized and degraded during bacterial life cycle for growth and division. PGN fragments act as messenger molecules in bacterial cell wall stress response and triggering mammalian innate immune response. Bacteria modulate their PGN degradation or recognition by chemically modifying PGN, such as amidating the epsilon-carboxyl group of meso-diaminopimelic acid in the PGN peptide chain. The quantitation of PGN-derived peptides is challenging due to their hydrophilic nature, but capillary zone electrophoresis coupled with high-resolution mass spectrometry shows excellent repeatability in quantitation of PGN peptides and their derivatives in bacterial cytoplasmic extracts.

In this study, capillary zone electrophoresis (CZE) -MS/MS was used to identify a large number of histone proteoforms with higher sensitivity compared to traditional methods.

Summary: The study reports the development of in vivo subcellular high-resolution mass spectrometry for proteo-metabolomic molecular systems biology in complex tissues. Through precision-translated fabricated microcapillaries, subcellular content of different cell types was analyzed with differential quantitative activities of proteins and metabolites identified. The technology preserved integrity of analyzed cells, neighboring cells, and embryos, with 95% of analyzed embryos developing into sentient tadpoles indistinguishable from wild-type siblings.
