Streptogramins for the treatment of infections caused by Gram-positive pathogens

Summary: Streptogramins, especially pristinamycin, can be considered as potential alternatives for the treatment of Gram-positive infections due to their rapid bactericidal activity and low prevalence of resistance among clinical isolates. Despite the effectiveness of quinupristin-dalfopristin in treating severe Gram-positive bacterial infections demonstrated in several studies, its clinical use remains limited mainly due to poor tolerance.

Summary: Longicatenamides A-D, new cyclic hexapeptides isolated from a combined-culture of two microorganisms, have their structures determined through spectroscopic analysis and absolute configurations of component amino acids determined using highly sensitive reagents. Particularly, Longicatenamide A shows weak but preferential antimicrobial activity against Bacillus subtilis.

Amycolapeptins A and B, Cyclic Nonadepsipeptides Produced by Combined-culture of Amycolatopsis sp. and Tsukamurella pulmonis

Summary: Two nonapeptide natural products with a 22-membered cyclic depsipeptide skeleton, beta-hydroxytyrosine, and a highly modified side chain were discovered in a combined-culture of rare actinomycete Amycolatopsis sp. 26-4 with Tsukamurella pulmonis TP-B0596. The planar structures were elucidated through spectroscopic analyses, and the absolute configurations of component amino acids were determined using the advanced Marfey's method. Unstable/unusual moieties in the structures were corroborated by chemical synthesis and CD analysis.
