NLRP3 inflammasome blockade reduces adipose tissue inflammation and extracellular matrix remodeling

Xabier Unamuno et al.

Summary: The study shows that NLRP3 plays a role in obesity and related metabolic changes, and blocking NLRP3 can alleviate AT inflammation with significant fibrosis reduction.

Hepatocyte pyroptosis and release of inflammasome particles induce stellate cell activation and liver fibrosis

Susanne Gaul et al.

Summary: The study found that hepatocyte pyroptosis, caused by NLRP3 inflammasome activation, leads to the release of inflammasome proteins and exacerbates fibrogenesis. Inhibition of caspase-1 and gasdermin D activation can prevent pyroptosis. The released inflammasomes are taken up by hepatic stellate cells, promoting inflammation and fibrosis. These findings suggest a novel mechanism for liver injury and potential therapeutic targets for chronic liver diseases.

Steatohepatitis Impairs T-cell-Directed Immunotherapies Against Liver Tumors in Mice

Bernd Heinrich et al.

Summary: This study found that steatohepatitis reduces infiltration of CD4(+) T cells and effector memory cells in liver tumors, which decreases the efficacy of immunotherapeutic agents like M30 and aOX40 in inhibiting tumor growth. N-acetylcysteine can restore T-cell numbers in tumors and enhance the ability of M30 and aOX40 to slow tumor growth in mice.

Summary: The global epidemic of obesity is associated with serious health consequences, including NAFLD, which can progress to liver fibrosis and cirrhosis. NAFLD is a systemic disease affecting multiple organs, with a complex pathomechanism involving PPARs as key regulators.
