Mixed myeloid chimerism and relapse of myelofibrosis after allogeneic stem cell transplantation

Samer A. Srour et al.

Summary: Assessing response and confirming early relapse after transplant for MF can be challenging, with detecting the JAK2 V617F mutation post-SCT being a strong predictor of relapse. Studies on the potential role of mixed chimerism, especially in patients with mixed myeloid chimerism, in predicting early relapse of MF are limited. This study aims to determine the role of myeloid chimerism in predicting relapse in MF patients and explore the correlation between myeloid chimerism and molecular relapse of MF.

Determinants of survival in myelofibrosis patients undergoing allogeneic hematopoietic cell transplantation

Juan Carlos Hernandez-Boluda et al.

Summary: This study aimed to evaluate survival determinants in myelofibrosis patients undergoing allo-HCT and describe factors predicting post-HCT complications. Factors associated with increased mortality included age, Karnofsky Performance Status, graft failure, aGVHD, and disease progression/relapse. Risk of aGVHD and relapse incidence varied with donor type and conditioning regimen, with GVHD having a complex impact on relapse and survival.

Impact of spleen size and splenectomy on outcomes of allogeneic hematopoietic cell transplantation for myelofibrosis: A retrospective analysis by the chronic malignancies working party on behalf of European society for blood and marrow transplantation (EBMT)

Nicola Polverelli et al.

Summary: The role of spleen size and splenectomy in predicting post-allogeneic hematopoietic stem cell transplant outcomes in myelofibrosis patients remains debated. However, splenectomy may improve survival and reduce non-transplant related mortality in patients with progressive disease and massive splenomegaly.

European wide survey on allogeneic haematopoietic cell transplantation practice for myelofibrosis on behalf of the EBMT chronic malignancies working party

Donal P. McLornan et al.

Summary: The survey revealed marked variations in practices across European allo-HCT centers for myelofibrosis, prompting future CMWP efforts to focus on defining guidelines for relapse and suggesting optimal monitoring practices for the transplant community.

Genomic analysis of primary and secondary myelofibrosis redefines the prognostic impact of ASXL1 mutations: a FIM study

Damien Luque Paz et al.

Summary: This study identified four genomic groups associated with the prognosis of myelofibrosis, with TP53 and high-risk groups showing significant associations with leukemic transformation and death from myelofibrosis. ASXL1 mutations alone were found to have no prognostic value but worsened the prognosis when associated with other high-risk mutations.
