Probing the Interaction of Huntingtin Exon-1 Polypeptides with the Chaperonin Nanomachine GroEL

Summary: Huntington's disease is caused by polyQ expansion in the exon-1 region of huntingtin protein, leading to aggregation and formation of inclusion bodies in neurons. The study shows that GroEL inhibits fibril formation of huntingtin protein and forms complexes with specific binding interface to sequester monomeric huntingtin, likely increasing the critical concentration required for fibrillization. The lifetimes of the complexes between huntingtin constructs and GroEL range from 100 to 400 μs with equilibrium dissociation constants of approximately 1-2 mM.
