Lipid-Raft-Targeted Molecular Self-Assembly Inactivates YAP to Treat Ovarian Cancer

Guanying Li et al.

Summary: The study introduced a molecular self-assembly technology that selectively halts cancer cell proliferation by inactivating the oncoprotein YAP. This technology targets the accumulation of ALPP and utilizes nanostructures specifically on lipid rafts of ovarian cancer cells to exert potent anti-proliferative effects.

Targeted Delivery of Persulfides to the Gut: Effects on the Microbiome

Kearsley M. Dillon et al.

Summary: This study presents a persulfide donor NDP-NAC that releases NAC-SSH in response to E. coli nitroreductase, leading to gastroprotective effects in mice and modulation of beneficial gut bacteria. It also reduces opportunistic pathogen populations in the gut, suggesting the potential of targeted delivery of reactive sulfur species for maintaining gut health or treating microbiome-related diseases.

Floxuridine Oligomers Activated under Hypoxic Environment

Kunihiko Morihiro et al.

Summary: This study reported the design and synthesis of floxuridine oligomer prodrugs, which are active under hypoxia conditions, can suppress tumor growth, and exhibit lower cytotoxicity. This is the first demonstration of conditional control of the medicinal efficacy of oligomerized nucleoside anticancer drugs.

Enzymatic Assemblies of Thiophosphopeptides Instantly Target Golgi Apparatus and Selectively Kill Cancer Cells**

Weiyi Tan et al.

Summary: Introducing a sulfur atom into phosphopeptides enables rapid targeting of Golgi apparatus and selective killing of cancer cells. The resulting thiophosphopeptide self-assembles after alkaline phosphatase-catalyzed dephosphorylation, enters cells, and accumulates in Golgi apparatus, inhibiting cancer cells effectively.

Supramolecular Antagonists Promote Mitochondrial Dysfunction

Mingming Li et al.

Summary: Mitochondrion-targeting therapy has great potential in cancer treatment, but limited therapeutic efficiency is a challenge. A mitochondrion-targeting supramolecular antagonist was developed, inducing tumor cell death by promoting apoptosis and preventing survival simultaneously. The combined therapeutic effect of the BH3 domain and CPT drug in the supramolecular antagonist efficiently inhibits tumor growth.

In situ oxidation-regulated self-assembly of peptides into transformable scaffolds for cascade therapy

Na Song et al.

Summary: The study demonstrates the oxidation-regulated self assembly of peptides into transformable scaffolds for cascade cancer therapy via in situ ROS generation. The scaffolds with combinatorial therapeutic effect promoted by photo-induced in situ ROS generation and morphological transition due to methionine oxidation enhance tumor accumulation and penetration of therapeutics, leading to effective tumor eradication. The findings provide a new and versatile strategy for ROS-associated disease treatment in the future.
