MnO2 decorated ZIF-8@GOx for synergistic chemodynamic and starvation therapy of cancer

The study developed a nanoplatform by decorating MnO2 nanodots, which showed impressive performance in killing cancer cells by combining chemodynamic and cell starvation therapy. This method utilizes oxidative stress therapy and starvation therapy on tumor cells, providing a new prospect for cancer treatment.
Recently, chemodynamic and cell starvation therapy have been regarded as an impressive method for the cancer treatment. Herein, a nanoplatform was developed by decorating MnO2 nanodots on glucose oxidase (GOx) encapsulated in ZIF-8, in which MnO2 nanodots can firstly consume overexpressed glutathione (GSH) to avoid the side reaction between GSH and center dot OH. Then, the MnO2 will release Mn2+ to trigger the beginning of Fenton-like reaction for the formation of center dot OH species by catalyzing H2O2, one of the oxidation products of D -glucose in tumor cells. Meanwhile, the GOx can degrade D-glucose, the essential nutrient for the tumor cells, to deliver H2O2, the precursor of center dot OH species, which will cause starvation for cancer cells. Vitro experimental measurement further demonstrated the prominent performance of the MnO2 decorated ZIF-8@GOx on killing cancer cells. We believe that the remarkable effect of chemodynamic and cancer starvation therapy in this work can provide a new prospect for cancer therapy.
