Using unnatural amino acids to selectively label proteins for cellular imaging: a cell biologist viewpoint

Summary: GFP revolutionized cell biology by enabling protein visualization in living cells, but newer imaging technologies face limitations due to size and photophysics of fluorescent proteins. An alternative approach of direct protein labeling with fluorescent dyes has been introduced, allowing for live-cell and super-resolution imaging without the need for protein-labeling tags.

Summary: Photoactivatable fluorophores are limited in their applications due to their size and the need for ultraviolet light activation. Research has focused on more efficient protein labeling technologies, including single-step labeling processes. By using genetic code expansion and replacing oxygen with sulfur within existing fluorescent amino acids, the photoactivatable fluorescent amino acid thioacridonylalanine (SAcd) has been successfully incorporated into proteins in a single step.

Dual film-like organelles enable spatial separation of orthogonal eukaryotic translation

Summary: Engineering new functionality into living eukaryotic systems by enzyme evolution or de novo protein design is a formidable challenge. By developing film-like synthetic organelles that support protein translation on the surfaces of various cellular membranes, we are able to equip eukaryotic cells with dual orthogonal expanded genetic codes that enable specific reprogramming of translational machineries with single-residue precision. The ability to spatially tune the output of translation within tens of nanometers not only has implications for understanding the function of membrane-associated protein condensation in cells, but is also important for synthetic biology.

Bio-orthogonal Red and Far-Red Fluorogenic Probes for Wash-Free Live-Cell and Super-resolution Microscopy

Summary: This study reports close proximity tetrazine-dye conjugates with minimal distance between tetrazine and the fluorophore, showing potential for live-cell imaging with features such as combination with unnatural amino acids, wash-free multicolor and super-resolution STED imaging, and SOFI imaging. These dyes pave the way for advanced fluorescence imaging of biomolecules with minimal label size.

Genetic encoding of a highly photostable, long lifetime fluorescent amino acid for imaging in mammalian cells

Summary: Acd is a fluorescent amino acid with high stability, quantum yield, and long fluorescence lifetime, superior to existing genetically encodable fluorescent amino acids for monitoring protein interactions and conformational changes. Engineered pyrrolysine tRNA synthetase mutants allow for efficient Acd incorporation in both E. coli and mammalian cells, with an optimal construct identified through comparison of protein yields and amino acid specificity. The use of Acd in FLIM provides strong contrast due to its long lifetime, compared to endogenous fluorophores and engineered fluorescent proteins with lifetimes less than 5 ns.
