BRET- and fluorescence anisotropy-based assays for real-time monitoring of ligand binding to M2 muscarinic acetylcholine receptors

Lukas Gratz et al.

Summary: This study compared BRET and FA fluorescence techniques in the characterization of ligand binding to GPCRs, using M2R and TAMRA-labeled ligands as a model system. The results showed that both methods yielded consistent affinities with radioligand competition binding experiments and provided real-time kinetic binding data. Additionally, the study demonstrated that BRET and FA assays can serve as valuable alternatives to radioactivity-based methods for screening purposes and for a precise characterization of binding kinetics.

Guanidine Derivatives: How Simple Structural Modification of Histamine H3R Antagonists Has Led to the Discovery of Potent Muscarinic M2R/M4R Antagonists

Marek Staszewski et al.

Summary: This study discovered a novel class of muscarinic receptor antagonists by making simple structural modifications to histamine H3R antagonists, some of which unexpectedly showed strong antagonist potency. The newly synthesized ADS10227 exhibited affinities of 2.8 nM and 5.1 nM at human muscarinic M-2 and M-4 receptors, respectively.

Budded baculoviruses as a receptor display system to quantify ligand binding with TIRF microscopy

Tonis Laasfeld et al.

Summary: A novel method utilizing total internal reflection fluorescence microscopy was developed for studying receptor-ligand interactions, combining G protein-coupled receptor display and immobilization on functionalized coverslips. This method offers high throughput, sensitivity, and detailed observation of the ligand-binding process, and can be extended for advanced assays integrating diverse detection modes.
