Tofacitinib Suppresses IL-10/IL-10R Signaling and Modulates Host Defense Responses in Human Macrophages

Summary: Single-cell RNA sequencing revealed upregulation of IFN-gamma production by CD41 T cells in GA lesions, which is associated with inflammatory polarization of macrophages and fibroblasts. Treatment with tofacitinib inhibited IFN-gamma, oncostatin M, IL-15, and IL-21 activity, leading to clinical and histologic disease remission in some patients and marked improvement in others. The Janus kinase-signal transducer and activator of transcription pathway is activated in GA, and Janus kinase inhibitors appear to be an effective treatment option.
